Abstract

An investigation has been done to study the effect of long-term oral doses vitamin c 100 mg/kgBW pretreatment on hypoglycemic effect of glibenclamide in spraguedawley rat. The research was purely one-way completely random experimental design. The subject were 30 rat divided to 5 groups, 1st group administered using CMC Na 0,5 %, 2sd group using glibenclamide 0,45 mg/Kg BW, 3rd using vitamin C 100 mg/kgBW once time in day for 1 day and glibenclamide 100 mg/kgBW, 4th group using vitamin C 100 mg/kgBW once time in day for 3 days and glibenclamide 100 mg/kgBW, and 5th group administered vitamin C 100 mg/kgBW once time in day for 7 days and glibenclamide 100 mg/kgBW. Before administered by glibenclamide the subject was fasted during 18 hours. The hypoglycemic effect of glibenclamide after concomitant vitamin C 100 mg/kgBW was measured following an oral glucose tolerance test (GOTT). All of subject was get glucose 1,75 g/Kg BW 20 minutes after administered by glibenclamide. The concentration of blood glucose was established in 0 minute before GOTT and 15, 30, 60, 90, 120, 180, and 240 minutes of blood glucose concentration using enzymatic method used GOD-POD reagent. The data were analyzed statistically with one-way ANOVA, following Tukey test (p<0,05). The result showed that long-term pretreatment vitamin C 45 mg/Kg BB days once time in day for 1, 3, and 7 was increasing hipoglychemic effect of glibenclamide. This was indicated by decreasing of AUC 0-240 (Area Under Curve) and increasing the percentage of hipoglychemic potency (p<0,05). Based on this result could concluded that longer pretreatment vitamin C 100 mg/Kg BW could increase hypoglycemic effect of glibenclamide significantly (p<0,05). keyword : Vitamin C, glibenclamide, hipoglychemic activity
